Bảo tàng được thành lập nhân dịp kỷ niệm 100 năm ngày sinh Chủ tịch Tôn Ðức Thắng (20/8/1888 - 20/8/1988) tại toà nhà vốn là tư dinh của Trần Thiệu Khiêm, Thủ tướng của chính quyền Sài Gòn trước năm 1975. Bảo tàng là nơi lưu giữ, nghiên cứu, trưng bày, giới thiệu, về cuộc đời và sự nghiệp của Chủ tịch Tôn Ðức Thắng - nhà yêu nước lớn, một chiến sĩ kiên cường mẫu mực. Chủ tịch Tôn Ðức Thắng là người Việt Nam duy nhất đã tham gia phản chiến trên chiến hạm France tại biển Ðen vào năm 1917, ủng hộ cuộc cách mạng vô sản đầu tiên trên thế giới thắng lợi - cách mạng tháng Muời Nga. Là người kế tục chức vụ Chủ tịch nước Việt Nam sau khi Chủ tịch Hồ Chí Minh qua đời, từ năm 1969 đến năm 1980.Hiện này, Bảo tàng có 07 phòng trưng bày với diện tích trên 700 m2.
