Để làm ra một bức tranh gạo đẹp có giá trị, có chất lượng tốt, người nghệ nhân phải rất tỉ mỉ trong quá trình trọn nguyên liệu làm tranh gạo vì nó sẽ ảnh hưởng trực tiếp đến chất lượng của bức tranh.
Nguyên liệu làm tranh gạo là những hạt gạo đều và chắc được cấy trồng và thu hoạch trên những cánh đồng của quê hương Việt Nam, hiện có 3 loại gạo được chọn làm chất liệu vẽ tranh. Trong đó gạo tẻ được chọn cho những chi tiết mảnh, gạo nếp chọn cho những chi tiết to, rộng, gạo tấm dùng để tạo những chi tiết nhỏ và làm mịn bức tranh. Hạt gạo sẽ được tạo màu sắc khác nhau từ trắng, vàng, nâu, đen bằng cách sấy, rang mà không sử dụng bất kỳ một loại hóa chất tạo màu nào khác, màu sắc của gạo sẽ phụ thuộc vào nhiệt độ và thời gian sấy/rang. Vậy nên tranh gạo chỉ có các màu cơ bản vừa nêu trên mà không có những màu sắc sáng màu như xanh, đỏ, hồng, tím…Hiện tại, màu sắc của gạo làm tranh trên thị trường rơi vào khoảng từ 15 – 20 màu.
