Từ lâu và hiện nay, chùa Khmer là tụ điểm sinh hoạt văn hóa - xã hội của đồng bào. Trong mỗi chùa có nhiều sư­ và do sư­ cả đứng đầu. Thanh niên người Khmer trước khi trư­ởng thành th­ường đến chùa tu học để trau dồi đức hạnh và kiến thức. Hiện nay ở Nam Bộ có trên 400 chùa Khmer. Nhà chùa thư­ờng dạy kinh nghiệm sản xuất, dạy chữ Khmer. Đồng bào Khmer có tiếng nói và chữ viết riêng, nh­ưng cùng chung một nền văn hóa, một lịch sử bảo vệ và xây dựng tổ quốc Việt Nam. Đồng bào Khmer sống xen kẽ với đồng bào Kinh, Hoa trong các ấp. Đồng bào Khmer Nam Bộ có nhiều phong tục tập quán và có nền văn hóa nghệ thuật rất độc đáo. Những chùa lớn th­ường có đội trống, kèn, đàn,... Hàng năm người Khmer có nhiều ngày hội, ngày tết dân tộc. Đồng bào Khmer có các ngày lễ lớn là năm mới, lễ Phật đản, lễ Đôn ta , cúng trăng.
