2.5. Đo mật độ tinh trùng
Xác định hệ số pha loãng: Quan sát số lượng tinh trùng trung bình trong một vi trường ở vật kính 40x để xác định hệ số pha loãng theo Bảng 2.1
Chuẩn bị buồng đếm: thấm một giọt nước lên hai gờ của buồng đếm Neubauer cải tiến, đặt lam phủ lên và ấn nhẹ cho lam được giữ chặt.
Cách pha loãng:
• Hút thể tích dung dịch pha loãng và tinh dịch theo quy định trong Bảng 2.6 cho vào ống eppendorf. Trộn đều bằng máy vortex.
• Hút 10 µl mẫu đã pha loãng cho vào buồng đếm. Để lắng khoảng 4 – 5 phút.
 
Cách đếm:
• Đếm các tinh trùng có đủ đầu và đuôi, không đếm các tinh trùng đầu kim.
• Đếm bắt đầu từ hàng thứ nhất của ô số 5 (ô trung tâm), đếm cho đến khi đủ ít nhất 200 tinh trùng và hết hàng thì dừng lại.
