Môi trường PDA sau khi được chuẩn bị và hấp tiệt trùng ta để nguội môi trường xuống khoảng 40-45oC (nhiệt độ này là thích hợp vì nếu nhiệt độ quá cao sẽ làm chết vi sinh vật và gây nguy hiểm cho người tiến hành thí nghiệm, còn nếu nhiệt độ thấp hơn thì môi trường sẽ bị đông) rồi đổ vào đĩa petri đã được cấy mẫu khoảng 15-20ml môi trường sao cho môi trường phủ hết bề mặt đĩa rồi xoay đều đĩa theo 2 chiều để phân tán đều dịch mẫu với môi trường nhằm tạo khuẩn lạc rời, sau đó để cho môi trường đông lại (khoảng 10-15p)
