Nội dung chính của phim trong mỗi tập, đều xung quanh việc một chú mèo tên là Tom cố đuổi bắt chú chuột Jerry để rồi kết quả là sự đổ vỡ trong nhà. Lý do để Tom đuổi bắt Jerry là những mâu thuẫn “trẻ con” và những ứng xử ngây thơ ngộ nghĩnh giữa chúng. kết thúc một tập phim, hầu hết Mèo thua do thói tự cao, và quan trọng nhất là do chuột tinh quái thông minh.
