Qui trình sắc kí cho phép tách một hỗn hợp các chất với sự có mặt của pha tĩnh và pha động. Trong sắc kí khí, pha động là khí, còn gọi là khí mang. Pha động sẽ chuyển hỗn hợp các chất qua cột tách với tốc độ dòng không đổi.

Chất hoạt động mạnh trong pha tĩnh thì sẽ di chuyển qua cột tách chậm, ngược lại chất hòa tan kém trong pha tĩnh thì di chuyển nhanh qua cột.
